Smetatinib is a small molecule MEK inhibitor (a mitogen activation inhibitor) that has been shown to control low-grade serous ovarian or peritoneal cancer.

Smetatinib, in previous studies, showed that sematantin also has the effect of treating metastatic biliary cancer.
The first-line treatment strategy for low-grade serous ovarian cancer is surgery followed by radiotherapy. However, this type of tumor grows slowly and responds poorly to traditional chemotherapy drugs. Because MEK gene mutations have been demonstrated in low-grade serous ovarian or peritoneal cancers, studies using MEK-1/2 protein kinases targeting mitogen-activated protein kinase (MAPK) pathway have been performed with seletinib. The study was performed on 52 patients with 100 mg of seletinib administered orally twice a day for 4 weeks, and 33% of patients received 12 or more cycles of treatment. Before the start of the study, 58% of patients had received 3 or more cycles of chemotherapy. Simetinib controlled disease in 81% of patients. In particular, 8 patients had complete or partial remission, and 34 had stable disease. The median progression-free survival was 11 months, and 63% of patients had a progression-free survival of more than 6 months. In addition, patients were well tolerated with simetinib and only 3 patients experienced grade 4 adverse events.
