Determination of fructose, glucose and sucrose by high performance liquid chromatography - refractive index method

At present, the determination methods of sugar in food mainly include chemical method, gas chromatography, liquid chromatography differential refractive index detection and the like. The chemical method is cumbersome and non-selective; the gas chromatography method requires derivatization because the sugar is non-volatile, and the operation is cumbersome; and because the ultraviolet absorption of the sugar is weak, it is not suitable to use the ultraviolet detector for detection. Therefore, the method is adopted in this paper. The refractive index detector and Comasil NH 2 column were used to separate fructose, glucose and sucrose. The experimental results show that the method has good separation and is suitable for the detection of fructose, glucose and sucrose.

Cometro High Performance Liquid Chromatography System;

Cometro (United States Connaught) 6000 LDI Pump ;

Scienhome (Tianjin Airlines) Comasil NH 2   2 50*4.6mm column ;

Mobile phase: acetonitrile - water = 70:30 ;

Flow rate: 1 ml/min ;

Injection volume: 15 μ L;

Second, the results and discussion

1. According to the spectrum of the reference substance and the mixed sample, the peak with a retention time of 10.178 min is fructose, the peak with a retention time of 12.347 min is glucose, and the peak with a retention time of 17.455 min is sucrose.

2 , using a refractive index detector, Comasil NH 2   2 50*4.6mm column separates fructose, glucose and sucrose samples with good peak shape and satisfactory resolution.
