iCheck® melamine quantitative quick check card (milk) instructions
(C/N: CS107-A)
Melamine ( MEL ) is a chemical raw material that should not be artificially added in foods and feeds. Long-term intake can cause damage to the reproductive and urinary systems, bladder and kidney stones, and can further induce bladder cancer. The Ministry of Health and other five ministries announced in the No. 10 announcement in 2011 that the limit value of melamine in infant formula is 1 mg/kg , and the limit of melamine in other foods is 2.5 mg/kg . Foods above the above limit are not allowed to be sold. The iCheck ® Melamine Quantitative Check Card measures the 0-500 ppb melamine content in milk .
2. Melamine quantitative quick check card test kit
1) iCheck® Melamine Quick Check Card ( 25 )
2) MEL micropores ( 25 )
3) Dilution Buffer CSB-4 ( 5 mL)
4) 1.5mL centrifuge tubes ( 25 )
5) 1mL tips ( 25 )
6) 200μL tips ( 50 )
7) MEL electronic calibration card ( 1 )
8) Operating instructions
3. Other materials required for the test
1) 200μL, 1000 μL micropipettes ( 1 each)
2) iCheck® quantitative detector ( with zero calibration sheet )
3) Centrifuge ( 7000 rev / min, or higher)
4) Timer
4. Preparation before testing and instrument calibration
1 ) Restore the quick check card, buffer and sample to be tested to room temperature for 30 minutes before use.
2 ) Equipment zero calibration: Insert the zero calibration piece into the quantitative quick test chamber and click “ Zero Correction †.
3 ) Equipment calibration: Insert the MEL electronic calibration card into the quantitative quick tester card slot, and click “ Read Calibration †. The electronic calibration card must not be removed during the measurement.
4 ) According to the number of samples, open the package, take out the quick check card and MEL micro-holes, and place the MEL micro-holes on the micro-well.
5. Sample processing and testing
1) A 1 mL sample of milk to a 1.5 mL centrifuge tube (if not detected immediately, can be stored refrigerated milk to avoid corruption, resulting in failure or pollution), 7000 rpm / min, centrifuged for 10 minutes.
2 ) Add 75 μL of the dilution buffer CSB-4 to the MEL microwell, then add 75 μL of the lower layer skim milk sample, pipette 5 times to mix, and take 100 μL to the sample hole of the quick test card.
3 ) After reacting at 37 °C ( ±2 °C ) for 10 minutes, immediately put the quick check card into the quantitative quick tester and click the “ analysis †reading, which is the actual concentration of the sample.
6. Result determination
1 ) Detection range: 0-500ppb, the limit of quantification is 50ppb .
2 ) This test card is a quantitative quick check card. If there is no red line on the control line ( C ), it indicates that the quick check card is invalid.
3 ) The measured value of melamine is greater than 80% of the internal or standard limit. Please re-measure it once. If it is still greater than the above value, please use the instrument method for confirmation analysis.
4 ) If the measured value is 500 ppb , dilute the sample with milk that has not detected melamine, and finally quantify the reading in the quick tester by the dilution factor, which is the actual concentration of the sample.
7. Precautions
1) The quick check card is used once during the warranty period. Store the refrigerator at 2-8 °C until it is balanced to room temperature before use.
2) Beware of the moisture in the quick check card, the aluminum foil packaging bag is damaged or the quick check card is damp, the test card should not be used.
3) Try not to touch the white film in the center of the quick check card.
4) Avoid direct sunlight during all incubations.
5) The small bag in the aluminum foil packaging bag is a desiccant and should not be taken into account.
6) Exceeding the shelf life cannot be used.
